My rear hub is a Novatec D462SB-SL-S5S-B12-11S, 32H, 148x12mm "Boost" and I am wanting to upgrade it to a Hope Pro 4 MTB Boost Rear Hub 32h 148x12mm boost

My question is will that hub fit with original spoke and rim?

I jus went on the fact that it's the same size as novatec although I could be wrong! Any help is appreciated

I believe Novitec has a larger spoke hole diameter. I bet you will need new spokes.
