Ragley Marley 1.0 Clunk

Hi on my Ragley Marley when I break there is a loud clunk or snap noise which also happens when I begin pedaling on the downstroke but only when standing up. The rear wheel is tight but I'm not sure what it could be. Rear hub maybe. Any help much appreciated Thanks
